原文摘要:The injectable self crosslinking mixed hydrogel composed of collagen I and thiolated hyaluronic acid can alleviate the contraction of collagen I in vitro and overcome the weak cell adhesion site of hyaluronic acid. Five groups of injectable hydrogels with different ratios were prepared, and their gel time, injection force, mechanical properties, swelling capacity and disintegration properties were studied. These results show that Col7HA-SH series 3 hydrogel achieves the best controllable and injectable effect, the gelation time is only 10 seconds under 3.5 N injection force, and the storage modulus of the hydrogel can reach 11 kPa at 10 Hz frequency. In addition, phenotype maintenance, biocompatibility, and chondrocyte proliferation were performed through CLSM, SEM, histological staining, immunohistochemical staining, MTT assay, and glycosaminoglycan quantification. Similarly, Col7HA-SH series 3 mixed hydrogel encapsulated chondrocytes showed the best proliferation potential, phenotype maintenance, biocompatibility and convenient operation characteristics. These findings may be close to the potential clinical application of mixed hydrogels in cartilage repair.

 

 

 

透明质酸(HA)和胶原蛋白是软骨细胞外基质的关键组成部分胶原蛋白作为软骨组织的组成部分,也是细胞外基质 (ECM) 的结构蛋白。Col I具有良好的生物相容性和生物降解性,还包含大量的细胞粘附位点。然而,仅由 Col I 制成的支架存在机械强度差、降解迅速和免疫原性等缺点。此外,Col I在与细胞长时间体外培养后往往会严重收缩。同样,HA也是ECM的重要组成部分,其中其结构和生物学特性介导其在细胞信号传导、伤口修复、形态发生和基质组织中的活动。HA 水凝胶具有良好的生物相容性并促进软骨形成,但由于缺乏细胞粘附位点,以及亲水和阴离子特性,它会抑制细胞粘附。鉴于Col I和HA各自的优缺点,研究一种HA-SH/Col I混合支架。以下是制备过程:

制备了 5 组不同配比的可注射水凝胶,研究其凝胶化时间、注射力、力学性能、溶胀能力和崩解性能。这些结果表明,Col7HA-SH 系列水凝胶达到了可控和可注射效果。此外,通过 CLSM、SEM、组织学染色、免疫组织化学染色、MTT 试验和糖氨基聚糖定量进行表型维持、生物相容性和软骨细胞增殖。同样,Col7HA-SH 系列混合水凝胶封装的软骨细胞表现出最良好的增殖潜力、表型维持、生物相容性和方便的操作特性。

结论:胶原蛋白 I 和硫醇化透明质酸组合(HA-SH/Col I)的可注射自交联混合水凝胶有助于维持软骨细胞的圆形或椭圆形表型,Col I 和硫醇化透明质酸的组合可以显着缓解 Col I 收缩的问题和硫醇化透明质酸上缺乏细胞粘附位点的问题。
